One of the most possible culprit will be Window Vista's new function, the User Account Control (UAC). It is a security function that prevents malware from receiving the privileges necessary to compromise the system. Click here to read more.

However, some programs and games can't seem to run with UAC turned on. Though not as safe, you can turn off UAC to run the program.

If this does not work, you can try to use run program as administrator. This can be done by right clicking the program icon and click on run program as administrator.
Another solution is changing the compatibility mode of the program to Window XP or older. To do this, right click on the program icon, click Properties, and click on the Compatibility tab.
There you go. Hope you have your program up and running.
